The goal of this study was to create a nano vesicular gel for transdermal distribution of Pioglitazone and to look into its anti-diabetic potential. Various combinations of soya phosphatidylcholine (SPC), edge activators, and pioglitazone were used to make transferosomes using the traditional rotary evaporation process (PZ). Vesicle size was 128.2nm, polydispersity index (PDI) was 0.107, zeta potential was 31.22mv, and drug entrapment efficiency (EE) was 75.99 percent in the optimized batch. It was then integrated into a Carbopol 934 base gel, which was tested for morphology, spread ability, in vitro and ex vivo release, and kinetics. Streptozotocin (STZ) induced anti-diabetic activity was tested. The drug loaded transferosomal gel exhibited 72.85% reduction in sugar level at the end of 12 hours, whereas the marketed formulation showed 67.63 percent reduction in sugar level at the end of 12 hours. Conclusion: The current work proved the ability of transferosomes to successfully deliver PZ and showed a significant drop in glucose levels after diabetes induction in wistar rats. The constructed technique demonstrated improved PZ delivery through the skin.
