Assessing the Nephrotoxic Impact of Oseltamivir in Male Albino Rats after Repeated Exposure

In order to determine its potential nephrotoxic effects, the effects of oseltamivir administration, effective against influenza viruses A and B, on some selected parameters of rat kidney function were investigated. Three classes were divided into eighteen (18) albino male Wistar rats with body weights ranging from 150-190 g. Results showed a significant increase (P < 0.05) in blood urea nitrogen (BUN), serum uric acid and serum creatinine in the T2 group receiving oseltamivir 1 mg/Kg.BW for six weeks, while the T1 group receiving oseltamivir at the same dose rate for just one week showed a significant increase (P < 0.05) in serum uric acid alone. Atrophy of the glomerular tufts, dilation of the Bowman space, deposition of hyaline droplets inside the lumen of the proximal and distal con-voluted renal tubules, cloudy swelling of the epithelial cells lining the renal tubules with perivascular lymphocytic cuffing and extreme obstruction of the interstitial renal blood vessels were shown by kidney histopathological lesions in T1 rats. Necrosis of the epithelial cells lining the renal tubules and cystic di-lation with total dissolution of their epithelial linings were seen in the kidneys of the T2 rats. Mild interstitial fibrosis was also observed and the Bowman’s capsule thickened. Similarly, with moderate medullary interstitial fibrosis, fibrous thickening of the kidney capsule was present. To conclude, repeated doses of oseltamivir in rats induced certain deleterious nephrotoxic effects in a time-dependent manner.
