Research on the Rise of Circulatory Endothelin (ET)- 1 and Endothelin Receptors (ETA, ETB) Expression in Kidney of Obese Wistar Rat

Context: Endothelin (ET)-1, a circulatory protein, and its receptors (ETA and ETB) have been reported to play a pivotal role in several diseases, including obesity, in different species. The research is intended to observe the extent of ET-1 circulatory and ETA/ETB expression in obese ventricles and kidneys compared to non-obese Wistar rats. Methods: obese groups at 14 and 34 weeks Wistar rats were compared at equivalent ages with non-obese controls. The ventricle and kidney circulatory ET-1, ETA and ETB concentrations were calculated by the Enzyme Associated Immunosorbent Assay (ELISA) technique following the conclusion of the two groups at weeks 14 and 24.

Results: The amount of circulatory ET-1, kidney and LDL expression of ETA and ETB in obese rats was significantly higher than that of control rats (T-test, P<0.05) in the elderly, with no discrepancies between ETA and ETB in the ventricle. No variations in circulatory ET-1, ETA and ETB expression levels were observed between the obese and younger rat control groups (P>0.05). HDL levels for both groups were below normal value. Conclusion: Obesity in obese elderly rats contributes to kidney vessel dysregulation due to ET-1 and ETA/ETB operation.
