Studies on Micropropagation of an Exotic Ornamental Plant, Calathea crotalifera, for Production of High Quality Plantlets

In two selected varieties of exotic ornamental plants, Calathea crotalifera, an effective protocol was developed for micropropagation. The effects of various sterilisation techniques, type of explant, and the combination and concentration of plant growth regulators on the induction of shoots were investigated. When treated with a combination of 30 percent (v / v) NaOCl, 70 percent (v / v ) ethanol, and 0.3 percent (w / v) HgCl2, axillary shoot buds sprouted from rhizomes in soil-free conditions showed a high induction rate of shoots with the lowest contamination percentage. The highest number of multiple shoots in the MS basal medium, supplemented with 3.5 mg / L of 6-benzylaminopurine (BAP), 1.0 mg / L of 1-naphthaleneacetic acid (NAA), 3% of sucrose and 6 g / L of plant agar for both varieties, was obtained in the present study and was used as a multiplication medium. When the young shoot bud explants were incised longitudinally before subculture, microshoots were strongly induced. In order to assess the effects of activated charcoal and L-glutamine on reducing the issue of necrosis, chlorophyll analysis has been studied. Compared to indolebutyric acid ( IBA), the maximum root induction on MS medium supplemented with 1.0 mg / L 1-Naphthaleneacetic acid (NAA) was registered. The complete regenerated plantlets were successfully acclimatised under greenhouse conditions in the soilless medium. This is the first rapid mass propagation study for C. All crotalifera.
